　几十年来，癌症患者的治疗手段主要有三种：化疗、放疗、手术治疗。但是这三种方法不仅疗效不理想，其副作用也很严重。因此，人们一直在找一种新的方式，既可以强有力的对抗肿瘤，又可以很好的控制副作用，并且在治疗结束后能长期保持对恶性肿瘤细胞的杀伤作用。

　关于增强免疫系统抵抗癌症能力的研究已经进行了数十年之久。早在19世纪末期，威廉?科莱就对癌症患者注射死细菌的混合液，他温和的加热溶液以杀死细菌并消除其危害，但混合液中仍有大量的细菌蛋白足以使患者机体出现免疫反应，以此来抵抗癌症细胞。

　但是，如果想继续提高癌症疫苗的特异性和疗效，还有许多问题尚待进一步解决：首先，患者体内的肿瘤为什么不能直接诱导机体的抗肿瘤免疫反应？这可能与肿瘤患者的免疫缺陷或患者T细胞信号传导障碍有关；还有，为什么肿瘤细胞可以逃避机体的免疫监视的？这可能由肿瘤细胞的MHC表达减低、转运抗原程序缺乏以及有些肿瘤产生的某种细胞素抑制局部或全身免疫反应所导致。另外，人类癌细胞成分复杂，肿瘤的抗原表达也不均一，所以想诱发患者的有效免疫反应，就可能需要用不止一种种肿瘤抗原免疫。
